I want to know how to password protect or encrypt a file on windows vista home premium, and if that same password or encryption will still apply in macintosh os tiger or leopard. I will be using a flash drive, if that makes a diference…

The ability to encrypt files and folders is not available in Windows Vista Starter, Windows Vista Home Basic, and Windows Vista Home Premium.
You will have to use a 3rd party software.

Use truecrypt http://www.truecrypt.org It works on both MacOS and any flavor of windows.
